I know how Heliophobe said it is a matter of timing when playing "samples" and if you are faking a waveform. Can these faked waveforms also work on title screens, or will it affect the responses to a push of a button to start a game?

You could easily fit a button-checking routine in there before the wait for the next hsync, as well as some color-cycling effects or some simple sprite animation. It depends how complicated you want to get with the sounds you're synthesizing, and how clever you are at sneaking in code between waiting for updates.
